dc.description.abstractWe determined the effects of AT1 and AT2 (selective no peptides antagonists angiotensin receptors), arginine vasopressin V1 receptor antagonist as well as L-arginine, a nitric oxide donor and NW-nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase inhibitor, injected into supraoptic nucleus (SON) on water and sodium intake induced by the injection of angiotensin II (ANGII). Male Holtzman rats weighing 200-250 g with canulae implanted into the SON were used. The drugs were injected in 0.5 μL over 30-60 sec. The water intake after injection of saline SAL+SAL 0.15 M NaCl was 0.40±0.1 mL 2 h-1; SAL+ANGII increase water intake. Losartan decreased the water intake induced by ANGII. PD123319 injected prior to produce no change in water intake induced by ANGII. AVPA prior to ANGII reduced the water intake with a less intensity than losartan. L-arginine prior to ANGII decreases the water intake at a same intensity than losartan. L-NAME prior to ANGII potentiated the dipsogenic effect of ANGII. Losartan injected simultaneously with L-arginine prior to ANGII blocked the dipsogenic effect of ANGII. These results confirm the importance of SON in the control of water intake and strongly suggest that AT1, V1 receptors interact with nitrergic pathways within the SON influencing the dipsogenic effect of ANGII.en
dc.description.abstractForam determinados os efeitos de AT 1 e AT 2 (Selective não péptidos antagonistas de receptores de angiotensina), arginina vasopressina V um antagonista do receptor, assim como L-arginina, um óxido nítrico dador e N W -nitro-L-arginina metil éster (L- NAME), um inibidor da óxido nítrico sintase, injetado no núcleo supraóptico (SON) na ingestão de água e sódio induzida pela injeção de angiotensina II (ANGII). Foram utilizados ratos Holtzman machos pesando 200-250 g com cânulas implantadas no SON. Os fármacos foram injetados em 0,5 μL por 30-60 segundos. A ingestão de água após a injeção de soro fisiológico SAL + SAL NaCl 0,15 M foi de 0,40 ± 0,1 mL 2 h -1; SAL + ANGII aumentam a ingestão de água. Losartan diminuiu a ingestão de água induzida por ANGII. PD123319 injetado antes para não produzir alteração na ingestão de água induzida por ANGII. O AVPA anterior a ANGII reduziu a ingestão de água com menos intensidade que o losartan. A L-arginina antes de ANGII diminui a ingestão de água na mesma intensidade que o losartan. L-NAME antes de ANGII potencializou o efeito dipsogênico de ANGII. Losartan injetado simultaneamente com L-arginina antes de ANGII bloqueou o efeito dipsogênico de ANGII. Estes resultados confirmam a importância do SON no controle da ingestão de água e sugerem fortemente que os receptores AT 1 , V 1 interagem com as vias nitrérgicas no SON, influenciando o efeito dipsogênico de ANGII.pt_BR
